Exploring a novel lung sealant based on functionalized polyoxazolines

    The lung is an intricate organ, comprising of tiny sacs that hold air, with a structure that somewhat resembles that of a sponge, being capable of holding up to five to six liters of air. Lung cancer still affects approximately two million people worldwide on a yearly basis, and surgery is often required. After surgery, about one in ten patients develop prolonged air leakage, which increases complications and mortality. Surgeons use a variety of techniques, including sutures, staplers and lung sealing devices (patches, sprays and glues) to repair injury to the lung. These lung sealing devices are promising for repairing lung injury, especially in diseased lung where sutures or staplers may cause tears in the lung, but currently are not sufficiently effective. This thesis investigates a novel lung sealing device, a patch based on functionalized polyoxazolines. First, studies are conducted to ensure methodological validity, including validation of air leak measurements, systematic review into animal models of air leakage and validation of a sheep model of acute air leakage. The novel lung sealant is then tested on ex-vivo porcine lungs, a sheep model of acute air leakage and a long-term biocompatibility model.
